ABSTRACT

 

In order to examine the effect of transporting temperature of ovaries from 

slaghterhouse to laboratory on the in vitro maturation, fertilization and 

development, transporting temperatures were classified into 4℃ group, 

10℃ group, 20℃ group and 30℃ group. Modified TCM199 was used for in 

vitro maturation of immature oocytes and development was induced by using 

chemically defined protein-free TLP-PVA. After oocytes collected from 

ovaries transported at different temperature were matured for 22-24 hours 

in vitro, the proportions of oocytes which reached MⅠ and MⅡ stages were 

independent of transporting temperature. When oocytes collected from 

ovaries transported at different temperature were fertilized in vitro, 

polyspermy was significantly increased in 4℃ and 10℃ group than in 20℃ 

and 30℃ group(p<0.01). Proportion of 2 cells stage in 20℃ group and 30℃ 

group were 72.3% and 74.5%, respectively. Those values were significantly

(p<0.01) increased compared to 4℃ group(12.9%) and 10℃ group(16.2%). 

Proportion of blastocyst stage in 20℃ group and 30℃ group were 14.2% and 

16.5%, respectively. Those values were significantly(p<0.01) increased 

compared to 4℃ group(0.6%) and 10℃ group(1.8%). It had no detrimental 

effect on in vitro development to reserve ovaries at 20℃ for 3 hours.
